NEW MATERIAL:The title plasmid derived from the microorganisms belonging to the genus Rhodococcus, ca.2.6 kb in size and having the following numbers of sites broken by restriction enzyme: (1) Sac I: 2; (2) BamH I: 1; (3) Pvu II: 1; (4) Sca I: 1; (5) Sph I: 1; and (6) Xho I: 1. USE:A vector for raising and modifying microorganisms. PREPARATION:For example, Rhodococcus rhodochrous ATCC 4276 is incubated in a medium followed by recovering the resultant microbes through centrifugation. The microbes are then suspended in a buffer solution and lysozyme is added to this solution followed by shaking the solution at 37 deg.C for 3hr, and an ethidium bromide-contg. buffer solution is added to the above solution. The resulting system is put to density gradient centrifugation and a plasmid fraction detected by ultraviolet irradiation is aliquoted, dialyzed and then gathered through ethanol precipitation. The fraction collected is then subjected to agarose electrophoresis, thus obtaining the objective cyclic plasmid. |
