The invention describes a procedure for the in vitro determination of allergy in vitro with the help of lymphocytes, so-called memory cells, in the blood, in which a lymphocyte fraction is isolated from defibrinated blood and is cultivated in the presence of an antigen with the aim of determination of eventual presence of memory cells for that particular antigen among the lymphocytes in which stimulated memory cells, so-called lymphoblasts, are determined by radioactive precursor incorporation into DNA and by morphological evaluation, which is characterized by the lymphocyte fraction which is repeatedly depleted of monocytes and other adherent cells. The antigen may be metals, organic and inorganic compounds, ceramics, glass ionomers, pigments and composite materials. The invention covers also the use of procedure for study of diseases which may be caused by metal allergy or in which metal allergy acts as promotor (contributing factor).
