PURPOSE:To prepare the objective protein having a correct structure by adding a reducing agent to as culture solution of the protein or to a supernatant of the culture solution to cleave an error S-S crosslinking bond, and subsequently oxidatively forming the correct bond of the protein. CONSTITUTION:In a reholding method for protain in which the wrong crosslink of a S-S bond in a different kind of protein secreted and produced by a recombination DNA method is again crosslinked into a correct S-S bond, a reducing agent is added to a culture solution containing the protein or to the supernatant of the culture solution to cleave the wrong crosslink, and subsequently subjected to an oxidation treatment to again crosslink the protein into a correct S-S bond. The target protein includes different kinds of protein secreted and produced by recombination DNA methods, such as human growth hormones, bovine growth hormones, interferon and hirudin. Examples of the reducing agent include mercaptoethanol, dithiothreitol, glutathione, cysteine, thioglycolic acid, mercaptoethanolamone, dithioerythritol, etc. |
