Plasmid having a heat-sensitive replication origin which has the following properties: a) it is capable of autonomous replication in Corynebacterium and of being maintained in Corynebacterium, and b) when a cell containing the plasmid is cultured between 31 and 37 DEG C, the replication of the plasmid is inhibited and at the same time the plasmid is eliminated from the cell, and process for achieving a homologous recombination in Corynebacterium by means of the plasmid.
