发明名称 CELLOBIOSE PHOSPHORYLASE AND PRODUCTION THEREOF
摘要 PURPOSE:To obtain cellobiose phosphorylase having such specific physicochemical characteristics as to catalyze the reactions producing D-glucose etc., by incubating specific bacteria belonging to Erwinia. CONSTITUTION:The bacteria belonging to Erwinia such as Erwinia sp. KT 531 are put to aerating culture under agitation in a liquid medium containing a carbon source such as cellobiose, nitrogen source such as ammonium sulfate, inorganic salt such as potassium primary phosphate and other nutrients. Thereby, the cellobiose phoshphorylase produced is chiefly accumulated in the microbes. Thence, the microbes collected from the culture solution are destructed by e.g. ultrasonic treatment followed by purification, thus obtaining the objective enzyme. The present enzyme has such characteristics as to catalyze the reactions producing alpha-D-glucose primary phosphoric acid and D-glucose and the reverse reactions thereof and ca.6 in the optimal pH value, etc. The present invention will lead to the new production technologies for cellobiose and other disaccharides.
