NEW MATERIAL:A gane coding a site-specific endonuclease SceI small subunit having a basic sequence of the formula. USE:For the preparation of a biochemical reagent, site-specific endonuclease SceI, useful for researching the structure and functions of a gene DNA. PREPARATION:For example, the preparation of the gene coding the site-specific endonuclease SceI small sub-unit of the formula is performed as follows; separating mitochondria DNA from a site-specific endonuclease SceI-producing yeast IAM4274, cleaving the DNA with a restriction enzyme EcoRI, subjecting the prepared DNA fragments to an agarose gel electrophoresis to transcript the fragments on a nylon film, screening the a gene coding the site-specific endonuclease SceI small sub-unit with a probe, recombining the screened DNA fragment into a plasmid, cloning the plasmid and subsequently determining the base sequence of the prepared plasmid DNA by a method where in the plasmid DNA is partially cleaved with a restriction enzyme. |
