NEW MATERIAL:A DNA containing a base sequence capable of coding a human FK506-binding protein having an amino acid sequence expressed by the formula. USE:For production of the human FK506-binding protein, etc. PREPARATION:A clone containing a human FKBPcDNA is isolated by screening using a DNA sequence synthesized, based on an amino acid sequence (Glu at the 31st residue to Asp at the 37th residue), of a reported bovine FK506-binding protein (hereinafter abbreviated to FKBP) as a probe from a human T cell cDNA library prepared by using)lambdagt11 manufactured by CLONTECH CO. as a vector. The resultant clone is then subjected to bacteriolysis to provide a phage, and phase DNA is then recovered therefrom and reacted with EcoRI in a buffer solution containing an RNase A to carry out agarose gel electrophoresis and southern blotting. The obtained clone has ECoRI fragments of about 1.5kb and is separated and purified by a glass powder method. |
