NEW MATERIAL:Peptidase TU-212 enzyme active to cut the proline-containing peptides or protected peptides of formula I at the sites shown by the arrows, inactive to decompose the (protected) peptides of formula II (Boc is 5- butyloxycarbonyl; Bz is benzoyl) and having the following properties. Optimum pH, 7.6-7.8 (stable pH is 6.6-8.5); optimum temperature, 60 deg.C (5min) and 37-40 deg.C (>=10min); stable up to 40 deg.C by 30min treatment and completely inactivated at 56 deg.C ; inhibited with ethylenediaminetetraacetic acid, o-phenanthroline, dithiothreitol and zinc ion; isoelectric point, pH8-9; molecular weight, about 65,000. USE:Production of peptide, analysis of peptide structure, etc. PREPARATION:A microbial strain belonging to genus Streptomyces and capable of producing TU-212 enzyme is cultured in a medium and the objective TU-212 enzyme is collected from the culture solution. |
