PURPOSE:To improve the transformation efficiency and structural stability by screening a plasmid derived from an actinomyces separated from soil of various places. CONSTITUTION:A plasmid derived from a strain belonging to the genus Micromonospora is selected to prepare plasmids having the following physico- chemical properties. (a) Plasmid p1418; Number of bases; 5.8kb. Sites sensitive to restriction enzymes; MluI, BclI, PvuII, XhoI and SmaI. (b) Plasmid pCYC24; Number of bases; 11.8kb. Sites sensitive to restriction enzymes; BglII, BalI, etc. (c) Plasmid pSG32; Number of bases; 11.7kb. Sites sensitive to restriction enzymes; KpnI, BamHI, etc., (d) Plasmid pKC47; Number of bases; 5.3kb. Sites sensitive to restriction enzymes; BamHI, etc. (e) Plasmid pRif33; Number of bases; 28.9kb, etc. (f) Plasmid p1240; Number of bases; 21.15kb, etc., (g) Plasmid p1434. (h) Plasmid pACT557.
