PURPOSE:To enable simple and accurate obtaining of activity in a short time by adding a physiologically active substance to mouse leukemic cell M1, producing and reacting the reduction activity with a tetrazolium compound. CONSTITUTION:A physiologically active substance (A) such as interleukine 6 is mixed with a cultured and activated mouse leukemic cell M1 (B). To the resultant mixture, is added 1-10mg/ml tetrazolium compound, such as 3-(4,5- dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide. Thereby, a reaction mixture solution (D) is obtained and then cultured at 36-38 deg.C under conditions of about 5wt.% CO2 concentration for 1-10hr to convert the ingredient (C) into a formazan compound (E). The aforementioned ingredient (E) is subsequently eluted with isopropanol, etc., and the absorbance of the resultant eluate is measured at 460-630nm wavelength to measure the activity of the physiologically active substance. |
