<p>PURPOSE:To simply and inexpensively produce an useful substance in good productivity by culturing an animal cell in a suspension state, adsorbing and separating the prepared useful substance with an affinity column, returning the elution flown down to the first process and subjecting to other processes. CONSTITUTION:A culture solution is charged to a culture tank 1 and inoculated with an animal cell such as a mouse hybridoma strain cell. While oxygen gas containing carbon dioxide gas is charged from a line 11, the culture solution is cultured with stirring at approximately 37 deg.C for about 10 days to prepare a culture solution having a desired concentration. The culture solution is passed through a pump P2 and a separation membrane device 2, and the culture solution containing an useful substance (e.g. IgG) is injected through lines 14 and 15 into an affinity chromatography column 3 which is preliminarily treated with adsorbing and desorbing buffer solutions, and maintained in an equilibrium state. The useful substance is adsorbed to the column 3, and the culture solution passed through the column 3 is returned into the tank 1 through a line 20. A buffer solution for adsorption is injected into the column 3 from a line 16 and flown out from a line 21 to wash the column 3, and a buffer solution for desorption is injected into the column 2 from a line 17 to recover the desorbed useful substance from the line 21.</p> |
