PURPOSE:To simply improve sensing power by adding anhydrous Na2SO3 and ferric citrate to a modified culture medium ingredient composed of peptone, soybean peptone, glucose, L-cystine, etc. CONSTITUTION:Peptone is blended with soybean peptone, glucose, NaCl, sodium thioglycolate, L-cystine, Na2SO3 and, as necessary, agar to provide a modified TGC culture medium (A). To 1l culture medium (A), is added 0.2-1.0g anhydrous Na2SO3. Filtered and sterilized citric acid in an amount of 0.3-0.7g is then added to produce a culture medium (B), for sensing sulfurization blackening bacteria. A desired bacteria sample is then inoculated into the aforementioned culture medium (B), and cultured to afford a blackened colony. Thereby, the sulfurization blackening bacteria of the genus Clostridium or Desulfotomaculum, such as Clostridium thermohydrosulfuricum or Desulfotomaculum nigrificans, are sensed by the aforementioned blackened colony. |
