<p>PURPOSE:To obtain a therapeutic agent for rheumatoid arthritis, etc., by immunizing a mouse with a cell of highly in ducing human B cell differentiating factor receptor (human BCDF-R) as an immunogen. CONSTITUTION:An antibody-producing splenocyte of Balb/c mouse immunized with human myeloma U266 cell suspended in sodium chloride injection is fused with myeloma cell X63-Ag8-6.5.3 of Balb/c mouse in the presence of polyethylene glycol to give a hybridoma FERM P-11406 (FERM P-11406) (A). Then the strain A is subjected to long-term continuous subculture in a medium containing hypoxanthine, etc., to give a culture mixture (B). The component B is purified with ion exchanger, etc., to give monoclonal antibody 1-39 having properties of inhibiting the binding of human B cell differentiating factor (human BCDF) to human BCDF-R, neutralizing biological activity of human BCDF, having 150,000 dalton molecular weight, E280nm=14.0 molar absorption coefficient and IgG2a as the kind of immunoglobulin.</p> |
