Detection or determn. of an amino acid sequence antigenically active within a known amino acid sequence of a protein or part of it comprises (1) synthesising of peptides each comprising a sequence of amino acids corresp. to a sequence within the known amino acid sequence. The peptides have overlapping amino acid sequences; (2) contacting the peptides with antibody against the protein or portion of interest; and (3) detecting or determining the presence or absence of antigen-antibody reaction between each peptide and antibody to indicate whether the peptide has antigenic activity.
