PURPOSE:To obtain the subject enzyme useful for the modification of oil and fat or for the synthesis of esters, etc., on an industrial scale at a low cost by integrating a DNA coding a mold lipase, culturing a yeast containing the obtained recombinant DNA in a medium and separating the mold lipase accumulated in the cultured product. CONSTITUTION:A chromosome DNA is collected by conventional process from Rhizopus niveus IFO 4759 strain (a lipase-producing strain) and treated with restriction enzymes. Each fragment is transformed to E. coli to prepare a gene library. The library is cultured and a transformant holding a DNA coding a mold lipase is selected by colony hybridization using the lipase cDNA of Rhizopus niveus as a probe. The DNA is extracted, bonded to a vector and inserted into a yeast to obtain a transformant. The transformed yeast containing the DNA coding the mold lipase is cultured in a medium and the lipase accumulated in the cultured product is separated therefrom. |
