A DNA fragment contg. a sucrose phosphorylase (SPL) gene of a mol. wt. of 1000 kD with a restriction enzyme sequence of HindIII, RsaI/ScaI, TaqI, EcoT141, RsaI, EcoRI, EcoRV and HindIII is claimed. SPL gene of (i) the amino acid sequence disclosed, (ii) the nucleotide sequence disclosed, (iii) a DNA sequence that hybridises with the disclosed nucleotide sequence and codes for a polypeptide with SPL activity, and (iv) a vector including a SPL gene which may be used for the prodn. of SPL. SPL is isolated from microorganism grown on a solid support or pref. grown in culture medium that may have one or several N sources. USE/ADVANTAGE - Method for prodn. of SPL (claimed), e.g., for industry.
