<p>PURPOSE:To more efficiently culture a callus tissue of a Hiba arborvitae genus plant in comparison with the conventional method by using a culture medium containing a C source, an inorganic component composed of a specified molar ratio of an ammonium nitrogen and nitrate nitrogen and plant hormones as the essential components. CONSTITUTION:On a culture medium containing a C source, an inorganic component and plant hormones as the essential components, a tissue of a Hiba arborvitae genus plant, preferably, e.g. Hondai Makino is cultured. In this case, the above used inorganic component must contain an ammonium nitrogen and a nitrate nitrogen in (1:4-8), especially (1:6) molar ratio. As the C source, hydrocarbons such as sucrose, derivatives thereof, etc., are exemplified and as the plant hormones, auxin, etc., is exemplified. A piece of callus excellently introduced from a piece of explant, especially petiole or lateral bud, cut from a Hiba arborvitae genus plant is cultured or subcultured on the above-mentioned culture medium. As a result of the above-mentioned culture, the above-mentioned callus tissue can be stably reproduced and grown without necrosis thereof.</p> |
