<p>Peroxidase enzyme is obtd. from horseradish. 4 vols. of supernatant of sprouted, soaked horseradish are mixed with ethanol so that the alcohol concn. should reach 80%.. The mixt. is allowed to stand for 24 hrs. at 4 deg. C.. The ppte. obtd. is solubilised at pH 7.4 with 0.15M TFS to which 0.10 pts. wt. of silicic-acid-magnesium salt are added. The ppte. obtd. is agitated for 24 hrs. with 0.5 vol. of 1M sodium-hydrogen-carbonate soln. contg. 15% acetone. The ppte. is washed with the same soln. and filtered. The filtrate is passed through a chromatographic DEAE-SEPHADEX A50 ion exchange column, which is equilibriated with 0.005 M TRIS-HCl buffer. Peroxidase is eluted with a gradient of 0.1-0.5M salt. in TRIS-HCl</p>
