PURPOSE:To obtain a thrombin-binding substance having a specific amino acid sequence in a large amount and in a low cost by culturing a transformant cell containing a specific recombinant vector and collecting a polypeptide from the produced cultured substances. CONSTITUTION:At first, DNA fragment having base sequence capable of coding amino sequence expressed by the figure is prepared. Then, a replicable recombinant vector containing said DNA fragment and a replicable manifestation vector, a transformant cell maintaining said recombinant DNA, and then a thrombin-bonding substance are prepared in order. The above-mentioned transformant cell is cultured and said thrombin-binding substance is separated by extracting from the cultured cell and/or culturing solution. Namely, a culturing solution containing thrombin-binding substance or extracted solution of the cultured cell is subjected to various column chromatographic purifications to afford the objective purified thrombin-binding substance. |
