<p>A hirudin or hirudin-like polypeptide having anti-thrombin activity is prepared by recombinant DNA methodologies. The hirudin or hirudin-like polypeptide is preferably expressed in E. coli or insect cells. The hirudin-like polypeptide is preferably a hybrid protein from HV1 and HV2 having the following amino acid sequence: HV12: Val-Val-Tyr-Thr-Asp-Cys-Thr-Glu-Ser-Gly-Gln-Asn-Leu-Cys-Leu-Cys-Glu-Gly-Ser-Asn-Val-Cys-Gly-Gln-Gly-Asn-Lys-Cys-Ile-Leu-Gly-Ser-Asp-Gly-Glu-Lys-Asn-Gln-Cys-Val-Thr-Gly-Glu-Gly-Thr-Pro-Asn-Pro-Glu-Ser-His-Asn-Asn-Gly-Asp-Phe-Glu-Glu-Ile-Pro-Glu-Glu-Tyr-Leu-Gln where the underlined sequence is the HV2 portion.</p> |
