PURPOSE:To obtain a useful medicinal composition by culturing human leukemia-derived cells. CONSTITUTION:K3T cells (Bikoukenkinki No.11275, FERM p-11275) are obtained by establishment from the peripheral blood of a human adult T-cell leukemia patient. The obtained cells are then transferred to an MEM medium containing fetal calf serum, insulin, a growth factor, etc., and cultured at 35-38 deg.C to obtain a cultured material and cells in the resultant cultured material are subsequently collected and transferred to an entirely synthetic medium containing no serum and subcultured to obtain a subcultured material. A supernatant separated from the subcultured material is then purified using salting out method, gel filtration chromatography method, etc., to produce the objective bioactive substance having the following properties: molecular weight 42000+ or -10000; isoelectric point 6.6+ or -0.4; growth promotion of human megakaryocyte-based cell strain. A medicinal composition can be prepared, as necessary, by using the above- mentioned active substance as the active component. |
