<p>PURPOSE:To improve accuracy and simplicity by using an mRNA extracted from a sample as a material and judging the type of a human leukocytic antigen with a probe labeled with an acridinium ester. CONSTITUTION:A probe in which an acridinium ester expressed by formula IV [R5 is lower alkyl, etc.; R6 and R7 are H, amino, etc.; R8 is formula V (p is an integer of 0-10)] is labeled through a linker arm expressed by formula I [X1 is S or amino; Y is formula II (X2 is halogen, etc.; R3 is lower alkyl, etc.), etc.; R1 is 1-10C aliphatic hydrocarbon chain; formula III is protecting group of X1; m is an integer of 1-3 which can be taken by R2; R2 is C, O, N, halogen, etc., having electron attractive group so that (R2)m-C- may protect X1; n is 1 or 2, provided that n is 1 when X1 is S and 1 or 2 when X1 is amino] is hybridized with a cDNA from an mRNA in a sample. Thereby, a difference of even one base can be discriminated.</p> |
