NEW MATERIAL:Purified Hinc II restriction enzyme produced by a recombinant. USE:A reagent for genetic engineering. PREPARATION:For example, Hemophilus influenza Rc is cultured in a culture medium, collected, suspended in a buffer solution, bacteriolysed by addition of lysozyme and subjected to extraction using a solvent for collection of chromosome DNA. The collected DNA is treated with a restriction enzyme and 3-7kp DNA fragments are subsequently recovered by the electrophoresis method. The recovered fragment is bonded to a vector to form a plasmid library and the resultant library is subjected to screening to select clones containing Hinc II-modifying enzyme gene. Plasmids are extracted from the above-mentioned bacterium and treated with the restriction enzyme to obtain Hinc II restriction.modifying enzyme gene. The resultant gene is bonded to a vector and inserted into a host microorganism for transformation and the obtained transformant is cultured, thus providing the objective Hinc II restriction enzyme. |
