发明名称 METHOD FOR SENSITIZING AND EMITTING LIGHT OF FIREFLY LUCIFERASE
摘要 PURPOSE:To enable obtaining of a luminous sensitizing technology with an ultrahigher sensitivity of firefly luciferase by the coexistence of a water-soluble polymer compound, surfactant or fluorescent substance with the firefly luciferin. CONSTITUTION:A required amount of firefly luciferin (D-luciferin) or an enzyme analog thereof is dissolved in an adequate amount of a reaction medium (a buffer solution) and luciferase which is a luminous enzyme, adenosine monophosphate (AMP), magnesium chloride, EDTA, a water-soluble polymer compound such as polyethylene glycol, various surfactants or fluorescent substances are added, dispersed and dissolved therein at 0 deg.C to ambient temperature. After the lapse of a prescribed time, a prescribed amount of the luminous liquid is taken out to measure the luminous quantity with a lumiphotometer and calculate amplification effects (increase rate %). As a result, a trace amount of a substance can be detected and measured with high sensitivity by using the aforementioned method for sensitizing and emitting light. Thereby, the method can be applied to methods for immunological measurement, measuring and detecting systems such as DNA probes or biosensors.
