PURPOSE:To enable to prepare pure deacetyl cephalosporin C acetyltransferase (DCPC ATF) having specific characteristics in a stable state by adding ATF BASE to a crude enzyme solution containing the DCPC ATF and subsequently subjecting the mixture to specified four step treatment process. CONSTITUTION:A mixture solution (hereinafter referred to as ATF BASE) comprising ethylene glycol, a sulfhydryl-containing reducing agent, 7-amino- cepholosporamic acid, EDTA and a serine protease inhibitor is added to a crude enzyme solution originated from a microorganism such as Acremonium chrysogenum (FERM 11232), and to mixture is subjected to the adsorption and elution of the enzyme with a weak anion exchange resin in the first step, with a hydrophobic chromatography in the second step, with a gel filtration in the third step, and with a weak basic anion exchange high performance liquid chromatography, thereby providing highly pure deacetyl cephalosporin C acetyltransferase (DCPC ATF) having specific characteristics on the amino acid sequence thereof, etc. |
