PURPOSE:To allow the easy and rapid detection of DNA which is amplified by a gene amplification method by executing the detection of the above- mentioned DNA by a capillary electrophoresis method. CONSTITUTION:The DNA is first extracted from the desired microorganisms, etc., in a specimen sample by a known method and this DNA is amplified by a PCR (Polymerase Chain Reaction) method. The PCR reaction liquid contg. the amplified DNA is directly analyzed by the capillary electrophoresis after the mineral oil in the upper layer is removed. The electrophoresis is effected by adding a polymer which is not gelatinized to a buffer soln. for electrophoresis in the capillary electrophoresis and any polymer is usable, insofar as the polymer is not gelatinized at the time of use. Such polymer includes, for example, the polymer known as low-melting agarose of a low gelatinization temp. |
