PURPOSE:To easily and quickly determine the number of yeast cells by adding quinone to a specimen containing yeast and measuring the generated H2O2. CONSTITUTION:A yeast-containing specimen (A) is prepared by diluting a cultured yeast liquid with imidazole:nitric acid, etc. A quinone (B) such as menadione is added to the specimen A and reacted at about 30 deg.C for about 3 min to obtain a reaction liquid (C). A luminous reaction liquid (D) containing bis(1,4,6-trichlorophenyl)oxalate, etc., is added to the liquid C to effect the color development corresponding to the amount of H2O2 generated in the component C. The number of living yeast cells can be determined by counting the developed color to obtain the amount of H2O2 and comparing the result with a preparatorily prepared calibration curve between the number of living yeast cells and the H2O2 productivity. |
