ANTI-NEOCARCINOSTATIN MONOCLONAL ANTIBODY AND HYBRIDOMA SECRETING THE SAME

PURPOSE:To obtain the subject substance utilizable in the determination and neutralization of an antitumor substance by culturing a specific hybridoma cell. CONSTITUTION:Human peripheral blood lymphocyte is transformed with Epstein- Barr virus(EBV) to obtain a B lymphoblast-like cell(BLC) (A) which is an antibody-secreting cell. Separately, mouse myeloma cell SP2 is cultured in the presence of about 1muM of ouabain to obtain a hybridoma parent strain (B) which is a ouabain-resistant strain. The strains A and B are mixed with each other at a ratio of (10-1):(1-10) and subjected to cell fusion in the presence of polyethylene glycol. The obtained fused cell (C) is cloned and the resultant hybridoma cell (D) is cultured to produce a human-type monoclonal antibody of human IgM-class antibody exhibiting antibody-recognition to neocarcinostatin and having heavy chain mu and light chain lambda.
