PURPOSE:To quickly and easily obtain a bacteriolytic component by separating bacterial cells from a suspension of specific bacteria with a filter, lysing the cells with a lytic liquid and extracting the obtained bacteriolytic component. CONSTITUTION:A specimen suspension liquid 1 containing campylobacter or Clostridium perfrigens is produced by dissolving human feces, etc., in a buffer solution. Separately, a filtering apparatus is prepared by assembling (A) a filter for removing coarse foreign materials and produced by laminating a silicone glass wool layer 7, glass fiber filter paper 8, etc., and (B) a holder 15 of a cell-supporting filter having a glass fiber filter paper 12, polyester filament mesh 14, etc. The above specimen 1 is put into a syringe 2 of the apparatus and sucked with a pump 16. After removing the holder 15 from the apparatus, a bacteriolytic liquid 20 is supplied to the holder 15 through a syringe 19 and cultured. The eluted liquid 21 is supplied to the cylinder 19 and sucked to recover the liquid in a suction bottle 22. The obtained bacteriolytic component is purified by extracting with phenol and precipitating with ethanol and the DNA in the bacteriolytic component is amplified by a polymerase chain reaction method. The presence of object bacterial cell in the specimen is determined by the amplified DNA. |
