PURPOSE: To provide a composition that contains individually specific amounts of tetracycline and non-steroidal anti-inflammatory agents and can treat mammals afflicted from tissue-destructive conditions relating to excess metalloproteinase activity. CONSTITUTION: This composition comprises (A) a tetracycline in such an amount that is effective as an anticollagenase but is not effective as an antibiotic substance, for example, 4-de(dimethylamino)-tetracycline, 6# a benzylthiomethylenetetracycline and (B) a non-steroidal antiinflammatory agent in such an amount that brings about marked reduction in bon destruction in combination with the component A in such an amount that is effective as an anticollagenase, for example, acetic acids, propionic acids, fenamates, pyrazoles or oxycums in amounts of A: 0.1-24mg/Kg/day and B: 0.3-3,500mg/kg/day. Rheumatoid arthritis, hypertrophic arthritis, corneal ulcer, epidermolysis bullosa, osteoporosis and other metabolic bone diseases, diseases bringing about damage to basement membrane, diseases accompanied by passing through the cells of the basement membrane and periodontal diseases are cited as curable diseases.
