NEW MATERIAL:Antibiotic substance RH5012-C of formula and its salt. It has following physical and chemical properties: white powder; water-soluble basic material; melting point, 191-194 deg.C (as free base); molecular formula, C20H37N3O13; molecular weight, 527; specific rotation,[alpha]D<20>=+12.9 deg. (C=1, water); solubility, easily soluble in water, hardly soluble in methanol, insoluble in ethanol, acetone, chloroform, etc.; color reactions, positive to ninhydrin reaction, anthrone reaction, and anisaldehyde sulfuric acid reaction, and negative to Sakaguchi's reaction. USE:Antimicrobial agent effective to Gram-positive bacteria, Gram-negative bacteria, and Candida albicans. PROCESS:The RH5012 strain (FERM-P No.5740) of actinomycetes belonging to Streptoverticillium genus separated from the soil collected at Shiga prefecture, Japan, is cultured in a medium containing untrient source to accumulate the antibiotic substance RH5012-C in the cultured product. The objective compound is obtained by separating the accumulated substance from the medium. |
