<p>PURPOSE:To safely prepare the subject antibody by administering a mouse- originated hybridoma cell M-5 strain (FERM 11035) into the abdominal cavity of a mouse. CONSTITUTION:Fused cells prepared by fusing myeloma cells with the splenic cells of a rat immunized with bilirubin oxidase are screened to prepare a hybridoma cell M-5 strain (A). The component A is administered into the abdominal cavity of a BALB/c-mouse, kept for approximately 10 days and subsequently subjected to the separation of the abdominal dropsy (B) of the mouse. The component B is treated to provide a monoclonal antibody (C) inhibiting the activity of the bilirubin oxydase. If necessary, the component C is microcapsuled or adsorbed and immobilized on a resin, etc., to provide a reagent (D) for analyzing vital components. The component D is compounded with a vital component sample such as blood and an oxidase such as glucose oxidase in the presence of the bilirubin oxidase, and the absorbance of a color caused by the generated H2O2 is measured to quantitate glucose, etc.</p> |
