<p>PURPOSE:To improve the diagnosis and therapy of human squamous cell carcinoma by culturing a specific hybridoma. CONSTITUTION:A BALB/C mouse is immunized with an immunogen containing a desired plasmid (e.g. pZipE 6.7), and a splenic cell separated from the immunized mouse is fused with a mouse myeloma cell, followed by cloning the fused cell to prepare a hybridoma 1NS-2 (FERM 11215) (A) producing a mouse monoclonal antibody 1NS-2 which specifically reacts with human squamous cell carcinoma. The component (A) is cultured in HAT medium (hypoxanthine- aminopterin-thymidine), etc., and the prepared culture supernatant (B) is separated and purified to prepare a 1gM type mouse monoclonal antibody which specifically reacts with human uterine cervix squamous cell carcinoma, human esophageal squamous cell carcinoma, etc.</p> |
