PRODUCTION OF CPB-I AND RECOMBINANT PLASMID AND TRANSFORMED YEAST USING THEREFOR

PURPOSE:To provide the subject plasmid which is a shuttle vector having a gene originated from yeast and a gene originated from E.coli plasmid, containing a specific anticoagulant substance (CPB-1) manifestation gene fragment and capable of producing CPB-1 in high efficiency. CONSTITUTION:The above recombinant plasmid is constructed of a shuttle vector having the above genes and has a CPB-1 manifestation gene fragment composed of Pho5 promoter, CPB-1-cDNA and GAP-DH terminator. The CPB-1-cDNA has been cloned by the present inventor and the details are disclosed in the specification of Japanese Patent Laid-Open Sho 64-20095. The CPB-1-cDNA is a gene fragment having a size of 1566bp and containing a gene fragment coding a peptide (CPB-1) consisting of 319 amino acids. It has been ascertained that the CPB-1 structure gene coded by the gene fragment codes the amino acid sequence of table 1.
