摘要

PURPOSE:To improve the productivity of Achromobacter collagenase by culturing a host cell transformed by a plasmid having a collagenase gene originated from Vibrio alginolyticus. CONSTITUTION:Whole DNA isolated from Vibrio alginolyticus capable of producing Achromobacter collagenase is partially decomposed with restriction enzyme Sau3A1 and the product is fractionated. Obtained DNA fragments of >=5kb are linked with a BamHI-treated vector pUC18 and T4 ligase and a host cell such as Escherichia coli JM101 is transformed to obtain a host cell transformed by a plasmid having a collagenase gene originated from a bacterial strain belonging to Vibrio alginolyticus. The host cell is cultured in a medium containing a carbon source, a nitrogen source and trace metal elements to obtain a collagenase.
