PURPOSE:To facilitate the separation of a target molecule by employing a signal-amplifying molecule comprising a signal region amplified as RNA and a target-combining region for forming a hybrid with the target molecule desired to be detected. CONSTITUTION:A specific plasmid DNA as a starting material is combined with an one chain DNA prepared by a known method to synthesize a two chain signal amplifying molecule comprising a promoter region which is the recognition combining site of a DNA-dependent RNA synthesizing enzyme, a signal region which is a transcription template and is amplified as RNA and a target-combining region for forming a hybrid with a target molecule desired to be detected. The detection of the target is performed by forming the hybrid of the target-combining region is the signal-amplifying molecule with the target desired to be detected, transcripting the signal region with a RNA-synthesizing enzyme, amplifying the signal region in a form of transcripted product and subsequently detecting the target. |
