PURPOSE: To measure the conjugation characteristics of a compd. to a natural receptor by conjugating a natural ligand or a cross reactive compd. with an enzyme donator segment ofβ-galactosidase. CONSTITUTION: A conjugant of a component capable of simulating the conjugation of a ligand to a natural receptor and aβ-galactosidase enzyme donator segment, a natural receptor and a compd. are mixed in an assay medium and aβ-galactosidase enzyme receptor segment is added to the medium to form holoenzyme along with the conjugant not bonded to the receptor. The enzymatic activity of the medium is measured as a measure of the conjugation of the receptor and the compd. The conjugant and an experiment compd. are allowed to compete with each other aiming at the receptor conjudgated with the receptor or an allosteric region to make it possible to determine the conjugation characteristics of the experiment compd. in relation to the enzymatic activity obtained when an enzyme receptor is added to the assay medium.
