PURPOSE: To form embryos of celery in high frequency and to cause germination from the embryos in high probability by growing callus cells of a plant in a maintenance medium contg. a low concn. of 2,4-dichlorophenoxyacetic acid (2,4-D). CONSTITUTION: In optional methods which are used for culturing embryos of celery and include a method that comprises growing callus cells of a plant in a maintenance medium contg. an about <2μm concn. of 2,4-D, the reproduction from the embryos is promoted by: (A) using only a low concn. of 2,4-D or no 2,4-D in a stage preceding the reproduction stage; (B) using a compound analogous to 2,4-D in the stage preceding the reproduction stage; (C) using sugar alcohol in the reproduction stage; (D) using sugar alcohol in an acclimation stage; (E) using abscisic acid in the acclimation stage; and/or (G) drying reproductive embryos.
