Disclosed are protoplasts of a plant of the genus Iris and methods of isolating and culturing same. The method of isolating the protoplasts of a plant of the genus Iris comprises transplanting lumps of tissue of a plant of the genus Iris induced in a plant tissue-culturing solid medium into a cell-culturing liquid medium to form fine and spherical cells, and converting the thus grown fine and spherical cells to protoplasts, and the method of cultureing the protoplasts comprises culturing the protoplasts in a liquid medium containing an auxin, a carbon source, and an osmotic pressure-adjusting agent.
