A process for culturing ginseng cell line ACG-2, KCTC 8974P (I) having much saponin comprises (A) gaining the ginseng stem, (B) removing remained strain by culturing (I) in Murashige-Skoog agar medium plus 3% sucrose and 0.1-3 mg/l 2,4-D for 4-5 wks at 15-25 deg.C with or w/o light. Cell mass of (I) at 0.1 mg/l of 2,4-D is less than in 3 mg/l 2,4-D, but with 1000 lux light, higher saponin as 2.89 mg/f lask.
