Activity-enhancing additives (I) for enzymatic reactions in which the substrate is a nucleotide-contg. mol. comprise milk and/or milk-components or milk prods. USE/ADVANTAGE - (I) are esp. useful for increasing the activity of restriction endonucleases in digesting nucleic acids, thereby reducing the amt. of enzyme required and rendering the digestion more complete (esp. important in studying restriction fragment polymorphism in genomic DNA). They may also increase the activity of ligases, ribonucleases, polynucleotide kinases, DNA and RNA polymerases, exonucleases and topoisomerases. (I) is dried milk powder or a protease-, DNase- and RNase-free milk protein fraction, esp. casein. (I) is added to an aq. buffer soln. of the enzyme and substrate in an amt. of 1-10,000 (esp. 50-300) mg/l. (I) may be formulated as a ready-to-use or concd. soln. in a buffer, pref. stabilised with a detergent and/or antifreeze.
