PURPOSE:To judge a number of samples within a unit time by a method wherein after a hydrogenation, grinding and centrifugal separation of a transformed plant cell having opine as marker, a filter paper is impregnated with a supernatant and placed on a filter paper for electrophoresis. CONSTITUTION:10-50mg of a plant tissue or the like regarded as being transformed is put into a sample tube. Water is added to the tissue or the like at a rate of 1mul per 1mg thereof, it is ground with a glass bar and separated centrifugally. A supernatant thus obtained is recovered into the sample tube. When 5-10mul of a sample liquid is applied onto a filter paper disc carried on a glass plate treated by silicon, the sample liquid is absorbed by the filter paper disc. The discs is placed at a sample applying position of a filter paper which is already immersed sufficiently into a buffer for electrophoresis and spread over a migration tank and is checked to see if the buffer permeates the discs. Migration is performed to confirm the presence of opine. Thus, the production of opine of a plant cell inoculated with agrobacteria can be judged quickly. |
