NEW MATERIAL:A hybrid promoter having a structure prepared by coupling a fragment containing an UAS of SUS2 promoter to a fragment containing TATA box site of GAP-DH promoter. USE:Used for production of human serum albumin(HSA) by the recombinant DNA technique. PREPARATION:For example, a SUS2 promoter-containing plasmid PY1011 is prepared from SUS2 gene and the resultant plasmid PY1011 is ligated to plasmid pUC18 to obtain plasmid pY1027. The obtained plasmid pY1027 is then coupled to a fragment prepared by digestion of plasmid PGG7 containing TATA box site of GAP-DH promoter using BglII-BamHI restriction enzyme and, furthermore, ligated to a fragment prepared by digestion of plasmid pYN026 using SphI-BamHI restriction enzyme, thus obtaining the objective hybrid promoter. |
