PURPOSE:To improve thermal stability and also durability without inactivating protein molecules even with a modifier at a higher concentration than that at which a conventional enzyme is inactivated and stabilize the subject molecules by enhancing hydrophobicity in the interior of the protein molecules. CONSTITUTION:Hydrophobicity in the interior of protein molecules is enhanced to stabilize the objective molecules. Further, the aforementioned molecules are Escherichia coli ribonuclease H and the 68th serine in the amino acid sequence of the natural type Escherichia coli ribonuclease H is preferably substituted with valine. Furthermore, a variant type Escherichia coli ribonuclease H structural gene coding the resultant stabilized variant type Escherichia coli ribonuclease H is preferably contained under conditions of controlled gene expression systems to provide a recombinant plasmid.
