<p>PURPOSE:To ensure adventitious embryos or callus to be induced by incubating saffron protoplast in such a state as to be embedded in an calcium alginate gel. CONSTITUTION:Firstly, cut pieces of sterilized saffron or its callus as raw material (cells) is subjected to pectiness- or cellulasebased enzyme followed by filtration and collection to obtain saffron protoplast. second, this protoplast is suspended in an aqueous solution of sodium alginate, and the suspension is dripped or extruded into an aqueous solution of calcium chloride to effect gelation of the aqueous solution of alginate in such a state as to contain the protoplast. Thence, auxin and cytokinin are added to an MS culture medium as the base medium, and the gel obtained above is drought into contact with or immersed in the culture solution followed by making an incubation. This results in vigorous proliferation to form cell aggregates. Thereby, incubation of the protoplast of crocus plants which has been unrealizable can be performed, and adventitious embryos or callus can be induced.</p> |
