PURPOSE:To enable simultaneous measurement of hemoglobin and leucocyte while obtaining same measured values as in the past by using a reagent containing no cyanogene while satisfying specified conditions. CONSTITUTION:This reagent is invented for calculation of leucocyte in blood and measurement of hemoglobin therein satisfying the following conditions and is characterized by containing no cyanogene: 1) The reagent must contain at least one kind of cationic surfactant selected from a group comprising compositions of a quanternary ammonium salt (formula I) or a pyridinium salt (formula I) which concentration is enough to dissolve red blood corpuscle in blood and to modify hemoglobin. In the formula, R1 is a8-20C alkyl group, alkenyl group or alkinyl group R2, R3 and R4 are respectively a 1-8C alkyl group, alkenyl group or alkinyl group, X<-> is an anion group and (n) is an integer of 7-19; 2) The reagent must contain at least one selected from a group compris ing both cationic and nonionic surfactants of the formulas III-V or an oxydizing agent which can oxydize hem in hemoglobin given by (f) and the like. In the formula, (f) represents nitrous ion, quinone body, alloxan or the like. |
