NEW MATERIAL:An anti-human tumor necrosis factor monoclonal antibody specifically reactive with human tumor necrosis factor. EXAMPLE:A clinical diagnostic for chronic, infectious and tumoral diseases. USE:The objective monoclonal antibody can be produced e.g. by diluting a human tumor necrosis factor prepared by gene recombination technique with physiological salt solution, mixing with Freund's complete adjuvant, immunizing a mouse BALB/c with the mixture by subcutaneous injection, extracting the spleen from the mouse 3 days after the final immunization, mixing with mouse myeloma cells to effect cell fusion, culturing the fused cell in HAT medium to obtain a hybridoma, screening the hybridoma to select a clone capable of producing an antibody specifically reactive with human tumor necrosis factor, cloning the clone e.g. by limiting dilution analysis and culturing the obtained monoclonized cell. |
