NEW MATERIAL:An endoserine recognition monoclonal antibody having an affinity constant Ka of >=1X10<10>M<-1>. USE:A reagent for the physiological and pathophysiological investigation of endoserine and a diagnostic for diseases such as hypertension and vasospasm. PREPARATION:The objective monoclonal antibody can be produced by linking endoserine to bovine thyroglobulin in the presence of a carbodiimide to obtain a complex, mixing the complex with Freund's complete adjuvant. immunizing a BALB/c mouse with the mixture by subcutaneous injection, etc., collecting the spleen cells after the final immunization, fusing the cell to mouse myeloma cell, culturing the fused cell in a hypoxanthine aminopterin thymidine culture medium to obtain a hybridoma, screening the hybridoma to select a clone capable of producing an antibody specifically reactive with endoserine, cloning the selected clone and culturing the obtained monoclonized cell. |
