A pretreatment for use in an assay for antigenic material in body fluids in which the body fluid is treated with a reducing agent comprising a compound containing an -SH group. The invention is particularly useful as a pretreatment for use with an assay for Class II antigens in body fluids (especially synovial fluids) of patients with rheumatoid arthritis.
