METHOD FOR UNSULATING AND CLEANING OF BETA- GALACTOSIDASE

This method is intended for use in the biotechnology industry. Using this method the enzyme is rapidly and effectively isolated from the biological bulk of Kluyveromyces Lactis and is cleaned without loss of its natural properties. This method is based on separation of the Kl. lactis cells from the fermenting liquid by micro filtration through membranes of 2.00 micrometer pore size at a pressure of 0.1 MPa. The cells are then suspended in a potassium/phosphate buffer of up to 5% dry weight and are destroyed by a homogenizing device, as a result of the high pressure of the flat head with a ballpoint operating at 65 MPa. The flow rate is 23.3 dm<3>/h, the temperature of the flow is 285 degrees K, which facilitates separation of the enzyme solution from the cell remains by filtration through a 0.4 micrometer pore size membrane under a pressure of 0.12 MPa. The substance is then subjected to cleaning and concentration using ultra filtration at a pressure of 1 MPa through 200 000 permeability limit membranes.
